Equipment of the Month: Exploris 480

The Proteomics Core Facility at the University of the Basque Country has installed a new Exploris 480 mass spectrometer coupled to an EASY nLC-1200 nanoUPLC system.

Description

This equipment is a state of the art hybrid quadrupole-Orbitrap mass spectrometer that combines high capacity transfer tube, high performance quadrupole analyzer with 0.4 Da FWHM precursor isolation widths and Ultra-high-field Orbitrap mass analyzer with a resolution up to 480000 (FWHM). Furthermore, the FAIMS Pro interface provides precursor ion selectivity based on differential gas-phase mobility that increases productivity and data quality. The new data acquisition platform supports multiple acquisition modes, such as DDA (Data Dependent Acquisition), PRM (Parallel Reaction Monitoring) and DIA (Data Independent Acquisition). Applications

This instrument can be used in most of the botton-up proteomics applications. The Exploris 480 can successfully perform botton-up type of analysis of low amount and/or high complexity samples.

  • Botton-up protein identification and quantification continues to be the basis for much of the proteomic research. It can analyze samples labeled with the newest isobaric mass tags as well as label-free ones.
  • Targeted quantification of known proteins with Stable Isotope Labeled (SIL) peptides.
  • Post Translational Modification analysis in order to determine the specific modification site and its relative quantification.

In the Proteomics Core Facility, we have been using quadrupole-Orbitrap mass spectrometers for more than 10 years and have a wide experience in the analysis of multiple kind of samples and different analytical workflows.
